In nonstick skillet, heat 1 tbsp of the oil over medium heat. Add onion, spinach, garlic powder, oregano and hot pepper flakes; season with salt and pepper. Cook, stirring often, until spinach is wilted, 3 to 5 minutes. Set aside.
Spread mozzarella on 2 of the tortillas, then spread reserved spinach mixture over top. Sprinkle with crumbled feta and top with remaining tortillas.
In large skillet, heat 1 tbsp of remaining oil over medium heat. Add one quesadilla to skillet; cook, turning halfway through cooking time, until cheese is melted and tortillas are browned, 6 to 8 minutes. Keep warm. Repeat with remaining oil and quesadilla. Serve with tzatziki, if using.
