In large saucepan of salted boiling water, cook pasta until al dente, 8 to 10 minutes. Reserving 1 cup of cooking liquid, drain pasta.
Meanwhile, in large skillet, heat oil and butter over medium-high heat. Add garlic; cook about 1 minute, stirring. Add mushrooms; cook, stirring occasionally, until golden, about 5 minutes. Add thyme and hot pepper flakes, if using; cook about 1 minute. Stir in cream, 1/2 cup of the Parmesan, the lemon zest and juice, and half of the reserved cooking liquid.
Add pasta, tuna and spinach to skillet, tossing gently to coat. Heat, stirring, until spinach has softened. Add remaining cooking liquid for creamier pasta. Season with salt and pepper. Sprinkle with remaining Parmesan; serve immediately.
